Robfine, while Dan's approach is undoubtedly the most rational in terms of seeking mutual buy-in and preserving a healthy relationship, here's a suggestion in case it's not enough and you need a credible fall-back: Indicate that you're going to go ahead and put spikes (perhaps even on gliders or seated in spike cups) under your speakers at their optimum location and then point out that moving the speakers will damage the floor.

Just the threat of that might be enough to resume negotiations to achieve a mutually acceptable solution that gets you to where you want to go. And note that you haven't threatened any tit-for-tat relocation or dislocation of her possessions in the process.
